Summary

We recently proposed intracellular signalresponsive gene regulation carrier system as a new strategy for cell specific gene delivery by using peptidegrafted polymer. In the design of the carrier, effect of polymer backbone, peptide pending moiety and other extra modified portions on the gene regulation ability and gene activation efficiency were investigated. The efficiency of gene regulation was evaluated by in vitro transfection and in vivo application. Finally obtained optimal polymer-peptide conjugate (protein kinase Cα- responsive) showed extremely high specificity to cancer cells. We also succeeded the stabilization of the polyplex in blood circulation.
